Identification of Avian Salmonella Isolates by PCR-RFLP Analysis of a fliC Gene Fragment
Authors
Abstract:
The genus of Salmonella is very polymorphic and comprised of a number of genetically closely related serotypes. It is one of the emerging pathogen in food-borne disease which is often found in contaminated chicken eggs. Salmonella enterica is considered one of the major pathogens in public health worldwide. A total of 31 Salmonella isolates identified by specific antisera, which included Salmonella enteritidis (51.6%), Salmonella typhimurium (25.8%), Salmonella infantis (19.4%) and Salmonella colindale (3.2%). DNA was extracted using phenol- chloroform- isoamylalchol method. All the isolates showed fliC gene (1500bp) by using specific primers. PCR products were subjected to digestion using HhaI restriction endonuclease. PCR- RFLP results showed 3 patterns between all isolates. Our research gained in this study demonstrated that using HhaI restriction endonuclease could differentiate Salmonella enteritidis and Salmonella colindale but there is similarity between pattern of Salmonella typhimurium and Salmonella infantis.

full textMolecular typing of avian Pasteurella multocida isolates from Iran by PCR-RFLP of OmpH gene
Molecular typing of twenty-five Pasteurella multocida isolates has been assessed by restriction fragment length polymorphism (RFLP) of a species-specific PCR assay. All of the P.multocida isolates were recovered from fowl cholera outbreaks in Northern provinces of Iran. Amplification was based on the gene ompH, encoding a major outer membrane protein (protein H). PCR with ompH primers amplified...
full textPCR–RFLP of isolated Salmonella from poultry with Sau3AI and HhaI restriction endonucleases in Arak
Salmonella enterica is considered as one of the major pathogens in public health, worldwide. Diseases caused by Salmonella enterica serovars are especially prevalent in developing countries. It is one of the emerging pathogen in food-borne diseases, which is often found in contaminated chicken eggs. The aim of this study was to apply a PCR–RFLP method using Sau3AI and HhaI restriction endonucle...
full textIdentification of gadoid species (Pisces, Gadidae) by PCR-RFLP analysis.
A rapid PCR-RFLP analysis was optimized to identify the presence of 3 closely related gadoid fish species: Alaska pollack Theragra chalcogramma, Pacific cod Gadus macrocephalus and Atlantic cod Gadus morhua in commercial seafood products. Gadoid universal primers were designed for PCR amplification of a 558-bp fragment encoding the mitochondrial cytochrome b gene. Without purification of the PC...

A rapid two-step identification scheme based on PCR-restriction fragment length polymorphism (PCR-RFLP) analysis of the 16S rRNA gene was developed in order to differentiate isolates belonging to the Campylobacter, Arcobacter, and Helicobacter genera. For 158 isolates (26 reference cultures and 132 clinical isolates), specific RFLP patterns were obtained and species were successfully identified...
